Impact of TP53 and E2F1 Gene Polymorphisms on Breast Cancer Prognosis

Pavadinimas: TP53 ir E2F1 genų polimorfizmų įtaka krūties vėžio prognozei. Tikslas: Įvertinti TP53 ir E2F1 genų polimorfizmų įtaką krūties vėžio prognozei. Uždaviniai:
- Nustatyti TP53 geno polimorfizmų (rs9895829 ir rs17884306) genotipų ir alelių pasiskirstymą krūties vėžiu sergančių pacienčių grupėje;
- Nustatyti E2F1 geno polimorfizmų (rs3213183 ir rs3213180) genotipų ir alelių pasiskirstymą krūties vėžiu sergančių pacienčių grupėje;
- Ištirti TP53 ir E2F1 genų polimorfizmų sąsajas su klinikinėmis ir patologinėmis naviko charakteristikomis;
- Ištirti TP53 ir E2F1 genų polimorfizmų sąsajas su krūties vėžio išgyvenamumu.
Metodika: Šiame tyrime dalyvavo 170 krūties vėžiu sergančių pacienčių. TP53 (rs9895829 ir rs17884306) ir E2F1 (rs3213183 and rs3213180) genų polimorpfizmai buvo genotipuoti naudojant genominę DNR, išskirtą iš pacienčių periferinio kraujo mėginių, taikant realaus laiko polimerazės grandininę reakciją (qPCR). Statistinė analizė atlikta naudojant „IBM SPSS Statistics“ programinę įrangą (30.0 versija) ir „Microsoft Excel”. Tyrimas buvo atliktas Lietuvos sveikatos mokslų universiteto Onkologijos instituto Onkologijos mokslo laboratorijoje ir yra laboratorijoje vykdomo tyrimo, kuris yra patvirtintas Kauno regioninio biomedicininių tyrimų etikos komiteto (protokolo Nr. BE-2-10 ir Nr. P1-BE-2-10/2014), dalis. Rezultatai ir išvados: Buvo išanalizuotas TP53 (rs9895829 ir rs17884306) ir E2F1 (rs3213183 ir rs3213180) genų polimorfizmų genotipų bei alelių pasiskirstymas tarp krūties vėžiu sergančių pacienčių. TP53 polimorfizmų genotipų ir alelių pasiskirstymas buvo toks: rs9895829 – AA (94.1%), AG (5.3%), GG (0.6%), A (96.8%) ir G (3.2%); rs17884306 – CC (92.9%), CT (6.5%), TT (0.6%), C (96.2%) ir T (3.8%). E2F1 polimorfizmų genotipų ir alelių pasiskirstymas buvo toks: rs17884306 – CC (92.9%), CT (6.5%), TT (0.6%), C (96.2%) ir T (3.8%); rs3213183 – AA (56.5%), AG (34.1%), GG (9.4%), A (73.5%) ir G (26.5%). rs3213180 – GG (82.4%), GC (15.9%), CC (1.8%), G (90.3%) ir C (9.7%). Statistiškai reikšmingų sąsajų tarp TP53 ir E2F1 genų polimorfizmų ir klinikinių bei patologinių naviko charakteristikų nenustatyta. Statistiškai reikšmingų sąsajų tarp TP53 ir E2F1 genų polimorfizmų ir išgyvenamumo be ligos progresavimo (PFS), išgyvenamumo be metastazių (MFS) bei bendrojo išgyvenamumo (OS) taip pat nenustatyta.
Title: Impact of TP53 and E2F1 Gene Polymorphisms on Breast Cancer Prognosis. Aim: To evaluate the impact of TP53 and E2F1 gene polymorphisms on breast cancer prognosis. Objectives:
- To determine the distribution of genotypes and alleles of TP53 polymorphisms (rs9895829 and rs17884306) in breast cancer patients;
- To determine the distribution of genotypes and alleles of E2F1 polymorphisms (rs3213183 and rs3213180) in breast cancer patients;
- To assess the associations between TP53 and E2F1 polymorphisms and clinicopathological characteristics;
- To evaluate the impact of TP53 and E2F1 polymorphisms on breast cancer survival. Methodology: A total of 170 breast cancer patients were included in this study. TP53 (rs9895829 and rs17884306) and E2F1 (rs3213183 and rs3213180) polymorphisms were genotyped using genomic DNA extracted from patients’ peripheral blood samples by quantitative real-time polymerase chain reaction (qPCR). Statistical analysis was performed using IBM SPSS Statistics software (version 30.0) and Microsoft Excel. The study was conducted at the Oncology Research Laboratory of the Institute of Oncology, Lithuanian University of Health Sciences, and is part of an ongoing laboratory study approved by the Kaunas Regional Biomedical Research Ethics Committee (protocol No. BE-2-10 and No. P1-BE- 2-10/2014). Results and Conclusions: The genotype distribution of TP53 rs9895829 was predominantly AA (94.1%), while AG (5.3%) and GG (0.6%) was less frequent. The A allele (96.8%) was more prevalent than the G allele (3.2%). Similarly, the genotype distribution of TP53 rs17884306 was mainly CC (92.9%), followed by CT (6.5%) and TT (0.6%). The C allele (96.2%) was more frequent than the T allele (3.8%). The genotype distribution of E2F1 rs3213183 was mainly AA (56.5%) followed by AG (34.1%) and GG (9.4%), while the A allele was more prevalent (73.5%) than the G allele (26.5%). For E2F1 rs3213180, the genotype GG (82.4%) was the most common, whereas GC (15.9%) and CC (1.8%) occurred less frequently. The G allele (90.3%) was more prevalent than the C allele (9.7%). No statistically significant associations were observed between TP53 and E2F1 polymorphisms and clinicopathological characteristics. Additionally, no statistically significant associations were found between these polymorphisms and progression-free survival (PFS), metastasis- free survival (MFS), and overall survival (OS).
