- Nustatyti RRP1B rs2838342 ir rs2051407 pasiskirstymą tiriamojoje kohortoje, įvertinant genotipo ir alelių dažnius.
- Įvertinti RRP1B rs2838342 ir rs2051407 sąsajas su klinikopatologinėmis charakteristikomis ir klinikiniais įvykiais.
- Įvertinti tiriamų polimorfizmų sąsajas su pacientų išgyvenamumu.
Metodika: Tyrime dalyvavo 191 pacientas, kurių DNR mėginiai buvo paimti Lietuvos sveikatos mokslų universiteto Onkologijos institute. Šiame tyrime RRP1B rs2838342 ir rs2051407 genotipavimas buvo atliekamas naudojant realaus laiko PCR. Visi anonimizuoti klinikopatologiniai duomenys buvo paimti iš anksčiau surinktų klinikinių įrašų. Statistinėms analizėms, įskaitant sąsajų ir išgyvenamumo analizes, atlikti buvo naudojama statistinė programinė įranga „IBM SPSS Statistics 30.0.0“. Tyrimo protokolą patvirtino Kauno regioninė biomedicininių tyrimų etikos komisija (protokolas Nr. BE-2-10 ir Nr. P1-BE-2-10/2014). Rezultatai ir išvados:
- Tyrimo kohortoje tiek RRP1B rs2838342, tiek rs2051407 polimorfizmų atveju dažniausiai pasitaikė heterozigotiniai genotipai (atitinkamai AG ir CT), o rečiausiai – homozigotiniai genotipai (GG ir TT). Alelių lygiu A alelis buvo labiau paplitęs nei G alelis rs2838342 atveju, o C alelis buvo dažnesnis nei T alelis rs2051407 atveju.
- RRP1B rs2838342 parodė, kad G alelis buvo susijęs su didesniu naviko dydžiu, o A alelis – su ER teigiamumu; be to, rs2051407 C alelis buvo susijęs su mažesne didesnių navikų tikimybe.
- Nebuvo nustatyta statistiškai reikšmingų sąsajų tarp RRP1B rs2838342 ir rs2051407 bei PFS, MFS ir OS.

- To determine the distribution of RRP1B rs2838342 and rs2051407 in the study cohort by assessing genotype and allele frequencies.
- To evaluate the associations of RRP1B rs2838342 and rs2051407 with clinicopathological characteristics and clinical events.
- To evaluate the associations between studied polymorphisms and patient survival.
Methodology: The study included 191 patients whose DNA samples were collected at the Institute of Oncology at the Lithuanian University of Health Sciences. In this study, genotyping of RRP1B rs2838342 and rs2051407 was performed using real-time PCR. All anonymized clinicopathological data were extracted from previously collected clinical records. Statistical software “IBM SPSS Statistics 30.0.0” was used to perform statistical analyses, including associations and survival analyses. The research protocol was approved by the Kaunas Regional Biomedical Research Ethics Committee (protocol No. BE-2-10 and No. P1-BE-2-10/2014). Results and conclusions:
- For both RRP1B rs2838342 and rs2051407 polymorphisms, heterozygous genotypes (AG and CT, respectively) were the most frequent in the study cohort, while homozygous genotypes (GG and TT) were the least frequent. At the allelic level, the A allele was more prevalent than the G allele for rs2838342, whereas the C allele was more frequent than the T allele for rs2051407.
- The RRP1B rs2838342 showed that the G allele was associated with larger tumor size, whereas the A allele was associated with ER positivity; additionally, the rs2051407 C allele was associated with a reduced likelihood of larger tumors.
- No statistically significant associations were found between RRP1B rs2838342 and rs2051407 and PFS, MFS, and OS.
