Šio darbo tikslas buvo patikrinti hipotezę ar alkilinančiais agentais, kaip TMZ, veiktos GBM ląstelės pakeičia savo transkriptomą taip, kad šiuos pokyčius būtų galima sieti su TMZ atsparumu. Pagrindinė darbo išvada – pasirinktose GBM ląstelėse atsiradę transkriptomo pokyčiai po periodinio TMZ poveikio, nėra tiesiogiai siejami su alkilinančių agentų reguliacija. Tačiau pastebėta, kad nustatyti neglobalūs transkriptomo pokyčiai gali būti siejami su ląstelės mechanizmais, kuriais pasirinktos GBM ląstelės gali netiesiogiai reguliuoti atsparumą TMZ. Šio darbo rezultatai prisideda prie gilesnio supratimo kaip GBM ląstelės, transkriptomo lygiu, reaguoja į TMZ poveikį. Tokio pobūdžio tyrimai reikalingi siekiant sukurti efektyvesnį chemoterapinį gydymą.
The aim of this work was to test the hypothesis that GBM cells exposed to alkylating agents, such as TMZ, alter their transcriptome in a way that could be associated with TMZ resistance. The main conclusion of the work is that the transcriptome changes that occurred in selected GBM cells after periodic exposure to TMZ are not directly associated with the regulation of alkylating agents. However, it was observed that the identified non-global transcriptome changes may be associated with cellular mechanisms by which selected GBM cells can indirectly regulate TMZ resistance. The results of this work contribute to a deeper understanding of how GBM cells, at the transcriptome level, respond to TMZ exposure. Such studies are needed to develop more effective chemotherapeutic treatments.
