Connexins form gap jungtion (GJs) that connect the cytoplasm of neighboring cells and unapposed/nonjunctional hemichannels that form a patway for the exchange of ions and metabolities between the cytoplasm and the extracellular milieu. Blocking hemichannels may enchance cell survival by preventing ionic imbalance, cell swelling and loss of critical metabolites. However, currently known hemichannel inhibitors also block GJ channels, thereby disturbing electrical and metabolic cell-cell communication. [...].
