Abstract
Magistrantūros baigiamojo darbo tyrimo tikslas – įvertinti pelėsinių grybų ir jų išskiriamų lakiųjų organinių junginių bei mikotoksinų įtaką kviečių, miežių, kvietrugių, avižų, rugių kokybiniams rodikliams sandėliavimo metu. 2019 metų derliaus grūdų mėginiai surinkti Pasvalio rajono X ūkyje rugsėjo, vasario ir liepos mėnesiais. Išdžiovinti grūdai sandėliuoti 5-7°C, sandėliuose buvo užtikrinta dezinsekcija ir deratizacija. Iškeltam tikslui įgyvendinti nustatytas bendras grūdų užterštumas pelėsiniais grybais (ksv/g), AFB1, ZEA, DON, OTA ir T-2 toksino koncentracijos (µg/kg) – plonasluoksnės chromatografijos metodu. Grūdų kokybinių rodiklių analizė atlikta spektrometru NIRS 6500. Be to, išorinį ir giluminį užterštumą sudarančios pelėsinių grybų gentys identifikuotos šviesine mikroskopija, o pelėsinių grybų riebalų rūgštys ir lakieji junginiai nustatyti dujų chromatografijos – masių spektrometrijos metodu.
Tyrimo metu nustatyta, kad vidutiniškai kvietrugių, kviečių, miežių, avižų rugių užterštumas pelėsiniais grybais, lyginant sandėliavimo pradžios ir pabaigos duomenis, padidėjo vidutiniškai 14,94 proc. (p<0,01). Mikotoksinų koncentracijos taip pat kito didėjančia tendencija, didžiausi pokyčiai AFB1 ir OTA kitime – koncentracijos atitinkamai padidėjo 18,67 ir 9,5 karto (p<0,01). T-2 toksino koncentracija padidėjo mažiausiai – 3,5 karto (p<0,01). Taip pat apibendrinus visų tyrime naudotų javų grūdų cheminę sudėtį, nustatyta, jog pelenų, ląstelienos, baltymų kiekiai mažėjo (p<0,01) – stipriausia neigiama, statistiškai reikšminga koreliacija – tarp T-2 toksino ir baltymų kiekio tyrimo viduryje. Riebalų (p≥0,05), drėgmės ir krakmolo (p<0,01) kiekiai didėjo, stipriausia teigiama koreliacija – tarp T-2 toksino, Rhizopus spp. paplitimo ir drėgmės. Grūdų pelėsinės taršos prevencijai buvo identifikuoti dažniausiai pasitaikančių mikromicetų lakieji junginiai – biomarkeriai. 3-etoksi-1,1,1,5,5,5-heksametilo-3-(trimetilsililoksi) trisiloksanas būdingas Alternaria alternata, 2,2,4,6,6-pentametilheptanas – Fusarium sporotrichioides, 2,5-dimetil-3-metileno-1,5-heptadienas – Cladosporium herbarum. Išvardintų mikromicetų sudėtyje dominuoja sočiosios riebalų rūgštys. 1-pentametildisililoksipropeną galima laikyti potencialiu Penicillium viridicatum biomarkeriu, minėtame mikromicete dominuoja polinesočiosios riebalų rūgštys. Aspergillus flavus, kurio sudėtyje vyrauja mononesočiosios riebalų rūgštys, iš kitų pelėsių išsiskiria 2,2,5,5-tetrametilheksano sinteze.

The aim of the Master‘s Thesis research work is to evaluate moulds and their released volatile organic compounds and mycotoxins’ effect on wheat, barley, triticale, oat and rye qualitative indicators during the storage. Grain samples of the year 2019 harvest in Pasvalys district X farm were collected in September, February and July. Dried grain was stored at 5-7°C, the store disinfestation and deratization were guaranteed. To achieve the set objectives, the total contamination of the grain with moulds (cfu/g) was determined, AFB1, ZEA, DON, OTA and T- 2 toxine concentration (µg/kg) identified using a thin-layer chromatography technique. Grain qualitative indicators’ analysis performed by NIRS 6500 spectrometer. Moreover, moulds tribes forming external and internal contamination were identified by light microscopy. Moulds fatty acids and volatile compounds were determined using the gas chromatography-mass spectrometry method.
During the study it was found that an average wheat, barley, triticale, oat and rye mould contamination, when comparing the beginning and end-of-storage data, increased by an average of 14,94% (p<0,01). Mycotoxins’ variation also had a growing trend; the most significance change was notices in AFB1 and OTA alteration where concentration increased accordingly 18,65 and 9,5 times (p<0,01). T-2 toxin concentration increased the least, it was 3,5 times bigger (p<0,01). The summary of chemical composition of all the grain varieties used in the study showed the reduction of ash, fibre and protein levels (p<0,01) – the strongest negative, statistically significant correlation between T-2 toxin and protein content in the middle of the study. Fat (p≥0,05), moisture and starch (p<0,01) levels increased; the strongest positive correlation between T-2 toxin, Rhizopus spp. spread and moisture. Biomarkers are the most common micromycetes volatile compounds to identify the grain mould contamination prevention. 3-Ethoxy-1,1,1,5,5,5-hexamethyl-3-(trimethylsilyloxy) trisiloxane characteristic to Alternaria alternata, 2,2,4,6,6-pentamethylheptane – to Fusarium sporotrichioides, 2,5-dimethyl-3-methylene-1,5-heptadiene – to Cladosporium herbarum. Saturated fatty acids prevail in the composition of the latter micromycetes. 1-pentamethyldisyloxypropene can be considered a potential biomarker of Penicillium viridicatum; polyunsaturated fatty acids dominate the said micromycete. Aspergillus flavus, containing predominantly monounsaturated fatty acids, differs from the other mould by synthesis of 2,2,5,5-tetramethylhexane.
