Liga pasireiškia motorinės, jutiminės ir kognityvinės funkcijos sutrikimais [1]. Šiuo metu IS nėra išgydoma, todėl gydymas orientuotas į paūmėjimų valdymą ligos eigą modifikuojančiais vaistais ir simptomų palengvinimą [2]. MiRNR – tai mažos nekoduojančios RNR molekulės, kurios reguliuoja įvairius ląstelės gyvybinius procesus, įskaitant ir remielinizacijos procesą, o kiekviena miRNR gali reguliuoti daugybės genų raišką. Šios molekulės yra centriniai imuninės ir CNS reguliatoriai, todėl yra pagrindinė autoimuninių ir neurologinių sutrikimų, įskaitant IS, tyrimų tema [1]. Vis dar ne iki galo aišku, ar skiriamas gydymas veikia dalyvaujančių remielinizacijos procesuose miRNR raišką kraujyje [3]. Tikslas: įvertinti remielinizacijos procese dalyvaujančių miR-219a-5p, miR-338-5p, miR-204-5p ir miR-17-5p raišką, atsižvelgiant į išsėtinės sklerozės gydymą. Medžiaga ir metodai: miR-219a-5p, miR-338-5p, miR-204-5p ir miR-17-5p molekulių raiška buvo nustatyta bendro kraujo mėginiuose pacientams iš keturių grupių, suskirstytų pagal iki tyrimo pradžios taikytą/netaikytą gydymą: 1 grupė – gydyti tik pirmos eilės ligos eigą modifikuojančiais vaistais (n=16); 2 grupė – gydyti tik antros eilės vaistais (n=38); 3 grupė – gydyti pirmos ir antros eilės vaistais (n=29); 4 grupė – pacientai, kuriems iki tyrimo pradžios nebuvo taikytas gydymas (n=10). MiRNR skyrimas buvo vykdomas naudojant „mirVanaTM miRNA Isolation Kit“ (Invitrogen, Masačusetsas, JAV) rinkinį. Kopijinės DNR sintezė atlikta naudojant „TaqMan Advanced miRNA cDNA Synthesis Kit“ (Applied Biosystems, Masačusetsas, JAV) rinkinį. MiRNR raiška buvo nustatoma tikro laiko PGR metodu, naudojant QuantStudioTM 3 System TL-PGR aparatą. Statistinei analizei atlikti buvo naudota GraphPad programa. Duomenų analizei apskaičiuotos miRNR molekulių ΔCt reikšmės, kaip referentinę miRNR naudojant miR-16-5p. Rezultatai: apskaičiuotos miR-219a-5p, miR-338-5p, miR-204-5p ir miR-17-5p ΔCt reikšmės. Jos buvo palygintos tarp tiriamųjų grupių: pacientų, kuriems iki tyrimo pradžios nebuvo taikytas gydymas, pacientų, gydytų pirmos eilės ligos eigą modifikuojančiais vaistais, pacientų, gydytų pirmos ir antros eilės vaistais, bei pacientų, gydytų tik antros eilės vaistais. Trijų iš keturių tirtų miRNR raiška statistiškai reikšmingai skyrėsi tarp pacientų grupių. Nustatyta, kad miR-338-5p molekulių raiška reikšmingai skiriasi lyginant pacientus, gavusius gydymą tiek pirmos, tiek antros eilės vaistais su pacientais, kurie buvo gydomi tik antros eilės ligos eigą modifikuojančiais vaistais (p=0,012). MiR-204-5p raiška statistiškai reikšmingai skyrėsi lyginant iki tyrimo pradžios negydytus pacientus su gydytais antros eilės vaistais (p=0.0006), gavusius gydymą pirmos eilės vaistais su gydytais antros eilės vaistais (p=0.0001) bei gydytus tiek pirmos, tiek antros eilės vaistais su gydytais tik antros eilės vaistais (p=0.034). MiR-17-5p raiška taip pat statistiškai reikšmingai skyrėsi tarp pacientų grupių. Reikšmingi skirtumai nustatyti lyginant pacientus, kuriems iki tyrimo pradžios nebuvo taikytas gydymas, su gydytais antros eilės vaistais (p=0.001), pirmos eilės vaistais gydytus pacientus su gydytais pirmos ir antros eilės vaistais (p=0.006), pirmos eilės vaistais gydytus pacientus su gydytais antros eilės vaistais (p<0.0001), gydytus pirmos ir antros eilės vaistais su gydytais tik antros eilės vaistais (p=0.001). Taip pat buvo įvertintos koreliacijos tarp tiriamųjų miRNR raiškos pacientų grupėse. Nustatytos dvi statistiškai reikšmingos koreliacijos: negydytų pacientų grupėje tarp miR-219a-5p ir miR-338-5p (r=0.77, p=0.021) ir gydytų tiek pirmos, tiek antros eilės vaistais pacientų grupėje tarp miR-204-5p ir miR-17-5p (r=0.49, p=0.009). Siekiant įvertinti miRNR diagnostinę vertę, buvo atlikta ROC analizė. Visos statistiškai reikšmingus raiškos skirtumus turėjusios miRNR, pasižymėjo didesniu nei 70% jautrumu ir specifiškumu (AUC>0,70). MiR-17-5p turėjo didžiausią diagnostinę vertę skiriant pacientus, gavusius pirmos eilės vaistus, nuo pacientų, gavusių antros eilės gydymą (AUC=0.9606). Išvados: miR-338-5p, miR-204-5p ir miR-17-5p molekulių raiška statistiškai reikšmingai skyrėsi tarp skirtingas ligos eigą modifikuojančias terapijas gavusių pacientų grupių, o jų gera diagnostinė vertė rodo potencialą būti IS gydymo poveikio ir remielinizacijos procesų biožymenimis. Nors nustatytas diagnostinis tikslumas perspektyvus, reikalingi didesnės apimties tyrimai šių miRNR klinikinei reikšmei patvirtinti.

Харків), 2026-04-15, p. 69-70Introduction. Exposure to environmental and lifestyle stressors (e.g., smoking, air pollution) can enhance oxidative stress, resulting in a higher need for antioxidants. Licorice (Glycyrrhiza glabra L.) roots are a potent source of natural antioxidants, primarily triterpenoid saponins and flavonoids, that exhibit antioxidant and anti-inflammatory effects. However, their biological efficacy is limited by poor stability and low membrane permeability. Liposome-based encapsulation may increase the bioavailability of these herbal constituents. Aim. This study aims to evaluate the effect of liposomes containing licorice (Glycyrrhiza glabra L.) root extract on reduced glutathione and malondialdehyde concentrations in the blood of mice. Materials and methods. The experiments were conducted on 4–6-week-old BALB/c laboratory mice divided into five groups (n=8): 1) control, 2) treated with aluminum solution 3) treated with liposomes containing Glycyrrhiza glabra L. root extract, 4) treated with aluminum solution and liposomes containing Glycyrrhiza glabra L. root extract, 5) treated with phospholipid suspension. Liposomes and phospholipid suspension (10 mL/kg) were administered orally via gavage. An aluminum solution (7.5 mg Al³⁺/kg) was delivered intraperitoneally to induce oxidative stress. Spectrophotometric analysis was used to determine reduced glutathione and malondialdehyde levels in the blood of mice after 21 days of daily exposure to the substances. Statistical significance was determined using one-way analysis of variance (ANOVA) and the unpaired Student’s t-test. The level of significance was taken as a value of p≤0.05. The data were analyzed using Microsoft Excel 2016 (Microsoft Corporation, Redmond, WA, USA). Results and discussion. Exposure to aluminum led to a statistically significant (p≤0,05) decrease in blood GSH concentrations and a significant (p≤0.05) increase in MDA concentrations compared to the control group. The results showed that liposomes containing Glycyrrhiza glabra L. root extract following aluminum exposure significantly (p≤0.05) increased reduced glutathione levels and significantly decreased (p≤0.05) malondialdehyde levels in the blood of mice compared to the aluminum-exposed group. Meanwhile, administration of liposomes containing Glycyrrhiza glabra L. root extract under normal physiological conditions had no effect (p>0,05) on the concentrations of reduced glutathione and malondialdehyde concentrations in the blood of mice. Conclusions. Treatment with Glycyrrhiza glabra L. root extract-loaded liposomes enhanced blood reduced glutathione and malondialdehyde levels under oxidative stress by aluminum, demonstrating their antioxidative activity.

Aim: The aim of this study was to assess the effect of liposomes containing Glycyrrhiza glabra L. root extract on brain glutathione (GSH) and malondialdehyde (MDA) concentrations in mice. Methods: Mice were divided into five groups (n=8): 1) control, 2) exposed to aluminum solution 3) exposed to liposomes with Glycyrrhiza glabra L. root extract, 4) exposed to aluminum solution and liposomes with Glycyrrhiza glabra L. root extract, 5) exposed to phospholipid suspension. Liposomes and phospholipid suspension (10 mL/kg) were administered by oral gavage, while an aluminum solution (7.5 mg Al³⁺/kg) was injected intraperitoneally to induce oxidative stress. Brain GSH and MDA levels in mice were determined spectrophotometrically after 21 days of daily exposure. Results: Aluminum exposure resulted in a statistically significant decrease in brain GSH concentrations and a statistically significant increase in MDA concentrations compared to the control group. In contrast, liposomes containing Glycyrrhiza glabra L. root extract significantly reversed the effects of aluminum by increasing GSH levels and reducing MDA levels compared with the aluminumexposed group, restoring both parameters to control values. Conclusion: Glycyrrhiza glabra L. root extract–loaded liposomes increased brain GSH levels and decreased MDA levels in mice under oxidative stress, indicating their antioxidative activity.

The expression of five miRNAs was analyzed in 88 participants: 45 in the myopia group and 43 in the control (emmetropic) group. Heatmap analysis revealed differences in the expression patterns of miR-29a, let-7i, miR-204, and miR-142 when comparing individuals with myopia and controls. The intensity of miR-328 expression was similar between the myopia and control groups. A significant correlation was observed between miR-204 ΔCt and spherical equivalent. However, comparison of median values between different degrees of myopia and the control group did not reveal significant differences. The simultaneous evaluation of multiple circulating miRNAs may help to better understand biological processes associated with myopia.

Glycyrrhiza glabra L. šaknyse daugiausia aptinkama triterpeninių saponinų ir flavonoidų, natūralių antioksidantų, kuriems būdingas ne tik antioksidacinis aktyvumas, bet ir kiti biologiniai veikimo būdai, tokie kaip uždegimą mažinantis, gastroprotekcinis, antimikrobinis poveikis [1]. Įkapsuliuojant į liposomas šias išorės veiksniams jautrias medžiagas galima pagerinti junginių bioprieinamumą bei pasiekti tikslesnį jų nukreipimą į atitinkamas organizmo vietas [2]. Tikslas. Įvertinti liposomų su paprastųjų saldymedžių (Glycyrrhiza glabra L.) šaknų ekstraktu poveikį redukuoto glutationo ir malondialdehido koncentracijoms pelių kepenyse. Metodai. Tyrimas atliktas su 4-6 savaičių amžiaus BALB/c linijos pelėmis (n=40). Pelės suskirstytos į 5 grupes pagal instiliuojamas medžiagas: 1) kontrolinę, 2) veikiamą aliuminio tirpalu, 3) veikiamą liposomų su Glycyrrhiza glabra L. šaknų ekstraktu, 4) veikiamą aliuminio tirpalu ir liposomų su Glycyrrhiza glabra L. šaknų ekstraktu, 5) veikiamą fosfolipidų suspensija. Oksidacinio streso sukėlimui naudotas AlCl3 tirpalas (7,5 mg/kg). Po 21 dieną trukusio tyrimo su pelėmis, nustatytos redukuoto glutationo (GSH) ir malondialdehido (MDA) koncentracijos pelių kepenyse taikant spektrofotometrijos metodą. Rezultatai. AlCl3 tirpalas statistiškai reikšmingai padidino MDA koncentraciją (p ≤ 0,05) ir statistiškai reikšmingai sumažino (p ≤ 0,05) GSH koncentraciją pelių kepenyse, lyginant su kontroline grupe. Statistiškai reikšmingas MDA koncentracijos sumažėjimas, lyginant su aliuminio tirpalu paveiktų pelių grupe, nustatytas pelių grupėje, kuriai buvo instiliuojamos liposomos su Glycyrrhiza glabra L. šaknų ekstraktu (p ≤ 0,05). Tyrimo metu taip pat nustatyta, jog liposomos su Glycyrrhiza glabra L. šaknų ekstraktu statistiškai reikšmingai sumažina oksidacinio streso metu padidėjusį MDA kiekį kepenyse (p ≤ 0,05), tačiau statistiškai reikšmingos įtakos MDA koncentracijai, lyginant su kontroline grupe, neturi (p > 0,05). Liposomos su Glycyrrhiza glabra L. šaknų ekstraktu statistiškai reikšmingai padidino GSH koncentraciją (p ≤ 0,05) pelių kepenyse, lyginant tiek su kontroline grupe, tiek su aliuminio tirpalu paveiktų pelių grupe. Ta pati tendencija pastebėta pelių grupėje, kuriai buvo girdomos liposomos su Glycyrrhiza glabra L. šaknų ekstraktu prieš tai sukėlus oksidacinį stresą aliuminio tirpalu. Išvados. Tyrimo metu nustatyta, jog liposomos su Glycyrrhiza glabra L. šaknų ekstraktu geba mažinti lipidų peroksidacijos žymens MDA koncentraciją ir padidinti GSH koncentraciją pelių kepenyse esant oksidaciniam stresui, o tai rodo galimą šių liposomų hepatoprotekcinį poveikį.

Identifying reliable biomarkers for disease progression and symptom severity remains a critical challenge. In this study, levels of oxidative stress-related microRNAs (miR-24-3p, miR-103a-3p, miR-320a-3p, miR-494-3p, miR-126-5p, and miR-543) within blood serum extracellular vesicles (EVs) were examined in a cohort of 93 PD patients to assess their associations with cognitive function, symptom severity, quality of life, and other clinical characteristics. The methods included microRNA extraction from blood serum EVs, followed by cDNA synthesis and RT-qPCR for expression analysis. Upregulation of miR-126-5p, as well as downregulation of miR-24-3p showed the strongest associations with symptom severity and cognitive decline, whereas downregulated miR-320a-3p levels correlated with patient-reported quality of life in PD patients. Downregulation of miR-103a-3p, and miR-543 expression showed slight associations with motor symptoms, cognitive function, and quality of life domains; however, some of these associations lacked statistical power. These findings indicate that specific microRNA expression profiles in extracellular vesicles are associated with PD symptom severity and progression, supporting their further evaluation as biomarkers in larger independent cohorts.

Ectracellular vesicles are small membrane-bound structures secreted by all types of cells and found in various biological fluids. Currently, the diagnosis of MS is based on clinical, radiological and laboratory criteria, but especially in the early stages, these markers are often non-specific. For this reason, additional diagnostic markers are being sought that can increase diagnostic accuracy and complement existing MS diagnostic methods. One of the significant molecules that can increase diagnostic accuracy is microRNA (miRNA). MiRNAs are small, non-coding RNA molecules that regulate gene expression and can be used as circulating biomarkers for various diseases, including MS. MiRNAs were isolated from serum extracellular vesicles from 49 MS patients and 49 controls using „ExoRNeasy Midi Kit“. MiRNA concentration and quality were assessed by using „NanoDrop 2000“ and cDNA synthesis was performed using the “TaqMan® Advanced miRNA cDNA Synthesis Kit“. MiRNA expression was detected by using RTPCR method. RT-PCR results were analyzed with „QuantStudio™ Design & Analysis Software“. The obtained data were processed and statistically evaluated using the „IBM SPSS Statistics 29.0.2.0“ program. It was found that the expression of both studied miRNAs was significantly different between the MS and control groups of neurologically healthy individuals (p0.05). ROC curve analysis revealed the diagnostic and prognostic potential of the studied miRNAs, assessed by AUC values. The results obtained indicate that the studied miRNA-21-5p and miRNA-106a-p may be suitable as potential biomarkers in multiple sclerosis.

The identification of reliable biomarkers is essential for improving our understanding of MS disease mechanisms and for developing more advanced diagnostic and prognostic strategies. microRNA expression profiling represents one of the most promising approaches for revealing molecular alterations associated with MS pathogenesis. The aim of this study was to identify potential MS biomarkers by applying sequencingbased expression analysis and machine-learning methods using untreated samples. MiRNA sequencing was performed on samples from 9 MS patients and 13 healthy controls. Following data quality control, principal component analysis (PCA) was applied to evaluate sample structure and group separation. Differential expression analysis was conducted using DESeq2, with results visualized through a volcano plot and heatmap. In parallel, sPLS-DA was employed to identify features contributing most strongly to group discrimination. Molecules overlapping between DESeq2 and sPLS-DA results were further evaluated using boxplots and univariate logistic regression models. PCA analysis showed a clear separation between the MS and control groups based on miRNA expression. DESeq2 identified miRNAs that were significantly differentially expressed. sPLS-DA further narrowed down the set of features with strong ability to distinguish the groups. All miRNAs selected by sPLS-DA overlapped with those found by DESeq2, supporting their stability and biological relevance. Logistic regression showed that even single miRNAs from this set could significantly separate the two groups. The integrated DESeq2 and sPLS-DA analysis identified a group of miRNAs that were statistically different between MS patients and healthy controls and showed strong discriminatory ability. The overlapping miRNAs represent strong candidates for potential diagnostic or prognostic MS biomarkers. The analysis was performed using untreated samples, making the results more reliable and providing a solid basis for future validation studies.

Its diagnosis and prognosis remain challenging – although clinical, radiological, and laboratory criteria are applied, reliable biomarkers capable of enabling earlier detection, predicting relapses, or assessing therapeutic efficacy are still lacking. Moreover, treatment effectiveness is often limited by inter-individual variability in therapeutic response and the complexity of disease progression. One promising class of biomarkers is microRNAs (miRNAs), small non-coding RNA molecules that regulate gene expression at the transcriptional or post-transcriptional level. However, due to their broad diversity, identifying biologically relevant miRNAs requires integration of in silico and experimental approaches. Our study consisted of two major stages: (1) in silico miRNA modeling and (2) an experimental sequencing phase. In the silico phase, we computationally derived predicted miRNA expression profiles in the context of MS. MiRNA sequencing was performed using the Illumina platform on extracellular vesicle samples isolated from blood (9 MS patients and 13 healthy controls). Differential expression analysis was conducted using the DESeq2 package to evaluate miRNA expression profiles between MS patients and healthy individuals. Overlapping miRNAs identified through both in silico prediction and sequencing were compared to assess their biological relevance to MS. Four miRNAs predicted in silico were confirmed in sequencing data. Among them, hsa-miR-19a-3p showed the highest expression change (log₂FoldChange > 1), while hsa-miR-17-5p, hsa-miR-139-5p, and hsa-miR-20a-5p exhibited moderate but significant alterations. KEGG analysis revealed enrichment in pathways of neurodegeneration – multiple diseases, amyotrophic lateral sclerosis (ALS), and the MAPK signaling pathway, suggesting shared molecular mechanisms between MS and other neurodegenerative disorders. The integration of in silico modeling with high-throughput sequencing analysis effectively identifies biologically relevant miRNAs in multiple sclerosis and highlights their potential as biomarkers involved in neurodegenerative and inflammatory signaling pathways.
