Aim: To determine whether hydrogen sulfide (H₂S) protects endothelial cells against hypoxia/reoxygenation-induced injury by reducing mitochondrial oxidative stress. Methods: Human umbilical vein endothelial cells (HUVECs) were exposed to 24 h hypoxia followed by 1 h reoxygenation in the presence or absence of the slow-releasing hydrogen sulfide donor GYY4137. Mitochondrial ROS production was assessed using MitoSOX Red and normalized to MitoTracker Green fluorescence. Expression of selected genes involved in endogenous H₂S synthesis, antioxidant defense and nitric oxide signaling was analyzed by RT-qPCR. Endothelial angiogenic capacity was evaluated using a Matrigel tube formation assay. Results: Hypoxia significantly increased mitochondrial ROS production compared with normoxic controls (P = 0.0077), and elevated ROS levels persisted after reoxygenation (P = 0.0386 vs. normoxia). GYY4137 markedly reduced mitochondrial ROS compared with both hypoxic (P = 0.0067) and untreated reoxygenated cells (P = 0.0334), returning mitochondrial ROS to near-normoxic levels. GYY4137 also increased the expression of genes involved in endogenous H₂S synthesis (CTH) and antioxidant defense (HMOX1), with HMOX1 expression significantly higher than in both hypoxic (P < 0.0001) and untreated reoxygenated cells (P = 0.0201). Despite these molecular changes, H₂S did not significantly improve endothelial tube formation after hypoxia/reoxygenation, indicating that restoration of redox homeostasis alone was insufficient to rapidly recover angiogenic function. Conclusions: H₂S reduced mitochondrial oxidative stress and restored the expression of cytoprotective genes following hypoxia/reoxygenation. Although these molecular effects did not translate into improved angiogenic function, they indicate that H₂S primarily targets early events in endothelial injury.

Introduction Atherosclerosis is a chronic inflammatory disease driven by interactions between immune responses and metabolic dysregulation [1]. While systemic inflammatory markers such as high-sensitivity C-reactive protein (hs-CRP) predict cardiovascular risk, local vascular inflammation also contributes to plaque development and instability [2,3]. Distinct systemic and local inflammatory pathways may differentially contribute to atherosclerosis and be reflected in circulating proteomic profiles. Aim To perform exploratory proteomic analysis in patients with non-obstructive coronary artery disease (CAD) and identify proteins associated with systemic and local vascular inflammation. Methods Eight patients with non-obstructive CAD confirmed by coronary computed tomography angiography were included. Blood samples were collected during the same visit. Perivascular adipose tissue (PVAT) fat attenuation index (FAI) was measured in the proximal 40 mm of the right coronary artery using Vitrea® software. Proteomic profiling covering approximately 11000 proteins was performed using the Illumina Somalogic platform. Protein expression differences were analyzed using DESeq2 in R. Correlations between FAI and protein levels were assessed with adjustment for CAD burden and risk factors. Results Patients with higher PVAT density showed reduced AHSG (fetuin-A) expression (fold change −1; p < 0.05) and increased PRR5L expression (fold change 1.94; p < 0.05). PVAT FAI negatively correlated with AHSG and positively with PRR5L. PRKRA expression was higher in patients with elevated hs-CRP but was not associated with PVAT FAI (p < 0.05). Conclusions Proteomic profiling identified proteins associated with PVAT-related inflammation in nonobstructive CAD. AHSG and PRR5L may reflect local coronary inflammation, while PRKRA may be linked to systemic inflammation. These findings support distinct roles of local and systemic inflammatory pathways in atherosclerosis. Larger studies are needed to confirm these results.

  • Matrix metalloproteinase 3 (MMP3), in conjunction with other MMPs, plays a key role in myocardial remodeling by breaking down the extracellular matrix of the heart and the development of heart failure (HF). However, the existing data on MMP3 protein expression in human myocardium remains insufficient. The objective of the present study was to determine the expression of MMP3 protein in left ventricular myocardial cells at varying stages of ischemic HF. To this end, a quantitative and semi-quantitative immunohistochemical analysis was performed on 113 samples of left ventricular myocardium. A non-selective digital myocardial image analysis revealed that the stage A HF group exhibited higher MMP3 protein expression compared to the control group (p < 0.001). Subsequent increases in immunostaining intensity of MMP3 were observed in the stage B HF group (p < 0.001). In the stage C/D HF group, MMP3 expression reached its highest level (p < 0.001). The intensity of immunostaining exhibited comparable tendencies in both cardiomyocytes and non-cardiomyocytes. Increased levels of MMP3 immunostaining in both types of cells were associated with increased left ventricular mass. Subjects who carried the 5A allele and patients with the 5A homozygous genotype exhibited a higher propensity for increased immunostaining levels in both cardiomyocytes and non-cardiomyocytes when compared to those with the 6A/6A and 5A/6A genotype. These changes in MMP3 protein expression were associated with left ventricular myocardial remodeling in the progression of ischemic HF.

The search for an effective treatment for adult high-grade glioblastoma (GBM) remains urgent. Background/Objectives: The study aimed to determine the expression of carcinogenesis-related genes, such as SLC12A2, SLC12A5, CDH1, CDH2, EZH2, and GFAP, in primary glioblastoma (WHO Grade IV; IDH-wild-type) cells from three adult women: GBM5-1, GBM5-2F, and GBM5-3F. Methods: The impact of the combination of sodium valproate and sodium dichloroacetate (2 mM NaVPA–3 mM NaDCA) on the expression of these genes was determined and compared with the effects of 50 µM temozolomide after 24 h of treatment. Results: 2 mM NaVPA–3 mM NaDCA, as well as temozolomide, had individual impacts on the SLC12A2, SLC12A5, CDH1, CDH2, EZH2, and GFAP expressions of tested GBM5-1, GBM5-2F, and GBM5-3F primary cells of female GBM patients. Conclusions: The combination of 2 mM NaVPA–3 mM NaDCA may have an advantage in antitumor activity and may be more effective than TMZ; however, the effect is individual.

Introduction. Cardioplegic solutions are employed during heart surgery to halt myocardial contraction. Their main purpose is to minimize myocardial oxygen demand by inducing electromechanical quiescence and cooling of the heart, thereby reducing ischemic effects. This process helps prevent degenerative changes, decreases the metabolic demands of the cardiac muscle, and protects cardiac myocytes from apoptosis and cell death [1]. Ischemic injury in the heart can be characterized by various molecular alterations within cells. BCL-2 family proteins play a crucial role in regulating mitochondrial apoptosis, either facilitating or preventing cell death [2]. In addition, FOS proteins have been associated with cell transformation, differentiation, proliferation [3]. The BNIP2 gene interacts with various proteins to protect cells from externally induced cell death [4]. Meanwhile, HIF1α serves as a key regulator of cellular and systemic homeostasis in response to hypoxia [5]. Aim. To assess the effects of three different cardioplegic solutions on immature rat heart tissues, we analyzed gene expression associated with the cell cycle, proliferation, apoptosis resistance, and hypoxia response. Methods. The animals were sacrificed, and the heart was removed through a median sternotomy, then transferred to a cold perfusion buffer. Within 5 min, the heart was mounted on a Langendorff system and perfused retrogradely. Cardioplegia was administered using St. Thomas (CP1), Custodiol HTK (CP2), or del Nido (CP3) solutions, with re-plegia intervals of 30 min (CP1), 90 min (CP3), or a single dose (CP2). As a positive control, fully perfused heart tissue was used (K-PRF). Ventricular biopsies were homogenized in liquid nitrogen to extract total RNA, which was then reverse-transcribed and analyzed for gene expression using TaqMan assays. Results. CP3-treated tissue had an average 50% lower BAX/BCL2 ratio than CP1 and CP2 tissue, however, these changes were not statistically significant. Following CP treatment, HIF1α gene was significantly overexpressed CP3 group compared to, CP1, and CP2 groups (P<0.05). In all cases, FOS expression was significantly decreased compared to the K-PRF after CP1 and CP2 treatment. After 2 and 4 h of incubation with CP3 solution, FOS gene expression remained substantially higher compared to CP1 (P<0.05) and CP2 (P<0.05). Similar results were observed in BNIP2 gene expression. Based on gene expression, CP3 treated cardiac tissue was comparable to K-PRF heart tissue in all timestamps. Conclusions. The CP3 solution exhibited the most effective cell-protective properties, comparable to heart perfusion. However, further studies are required to assess the electrophysiological and metabolic changes in heart tissue.

Introduction Cardioplegic solutions are used to stop myocardial contraction during heart surgery. Their primary goal is to reduce myocardial oxygen demand by creating electro-mechanical quiescence and cooling the heart, which reduces ischemic effects. This helps prevent degenerative changes, lowers the metabolic demands of the cardiac muscle, and prevents apoptosis and cell death in cardiac myocytes. [1]. Various alterations in molecular processes within cells can be used to define ischemic injury in heart. BCL-2 family proteins are important modulators of mitochondrial apoptosis, either promoting or inhibiting cell death [2]. Cell transformation, differentiation, and proliferation have all been linked to FOS proteins [3]. By interacting with various proteins, the BNIP2 gene protects cells from exogenously induced cell death [4]. HIF1α acts as a master regulator of the cellular and systemic homeostatic response to hypoxia [5]. Aim To evaluate three different cardioplegic solutions in immature rat heart tissues, we measured the expression of genes related to the cell cycle, proliferation, apoptosis resistance, and hypoxia response. Methods Animals were sacrificed, and the heart was excised via median sternotomy, transferred to cold perfusion buffer, and within 5 minutes, the heart was mounted on a Langendorff system. Cardioplegia was administered using St. Thomas (CP1), Custodiol HTK (CP2), or del Nido (CP3) solutions, with re-plegia intervals of 30 minutes (CP1), 90 minutes (CP3), or a single dose (CP2). The control group was regarded as non-CP tissue (CP0). As a positive control, fully perfused heart tissue was used (K-PRF). Ventricular biopsies were homogenized in liquid nitrogen in order to extract total RNA. RNA was reverse-transcribed and analyzed for gene expression using TaqMan assays. Data were normalized to GAPDH gene and analyzed with GraphPad Prism 9. Results CP3-treated tissue had an average 50% lower BAX/BCL2 ratio than CP1 and CP2 tissue, however, these changes were not statistically significant. Following CP treatment, HIF1α gene was significantly overexpressed CP3 group compared to CP0, CP1, and CP2 groups (P<0.01). In all cases, FOS expression was significantly increased compared to the CP0. Conclusions The CP3 solution demonstrated the best cell-protective properties, comparable to heart perfusion. However, further research is needed to evaluate the electrophysiological and metabolic changes in heart tissue.
