Abstract
Introduction: Glioblastoma multiforme is one of the most common astrocytomas. Despite surgical, radiotherapeutic and chemotherapeutic treatments, the average length of survival following diagnosis is approaximately 15 months. Histone deacetylase inhibitors attracted interest in most recent years due to increasing evidence that these inhibitors prove useful in cancer treatment. One of them - sodium valproate - is used in epilepsy, bipolar disorder and neuropathic pain treatment. The aim of this study was to evaluate the impact of using sodium valproate on tumors, created by U-87 glioblastoma cell culture, by using the chick embryo chorioallantoic membrane model. Materials and methods: During the course of this study, U-87 cells, treated with sodium valproate (4mM and 8mM), were planted upon chick embryo chorioallantoic membrane (CAM) on 7 day of embryo development (ED7) using absorbable gelatin sponge of 3mm³ and the process of tumor formation was observed. After five days (ED12) of incubation of tumors CAM's were cut and histological and immunohistochemical analysis of tumors (n=5-8), which formed on the membrane, was done. To evaluate neo-angiogenesis of untreated and sodium valproate treated groups (n=18-10) the mean number and area of blood vessels were counted. [...].
